Methods established in the laboratory:

14C-Fluorodeoxyglucose-autoradiography:

coronal section through the forebrain of an acoustically imprinted chick during exposure to the acoustic imprinting stimulus.

Immunohistochemistry:

in vitro:

double-Immuno staining,

rat, primary culture (14 days).

confocal laserscan microscopy:

filled LY-pyramidal cell, 3D-reconstruction

(Octodon degus).

Immunohistochemistry

14C-Fluorodeoxyglucose-autoradiography (analysis of stimulus evoked metabolic brain activity)

3H receptor-autoradiography: ligand binding for transmitter-peptide and hormone receptors.

quantitative light microscopy, confocal laserscan microscopy, e.g. immunocytochemical detection and ultrastructural localization of transmitter receptors, transmitter, Ca-binding proteins, synapse-specific proteins, enzymes

quantitative electron microscopy

in situ hybridization techniques

iontophoretic and pressure injection of fluorescent dyes in living and fixed neurons for three dimensional reconstruction and quantitative morphological analysis of dendritic trees and dendritic spines

computer-assisted image analysis for densitometry of autoradiographs, 3D-reconstruction of neurons (measurement of soma size, dendritic ramification patterns, density and spatial arrangement of dendritic spines), 3D-measurements of spine morphology (analysis system developed in our lab) and quantification of ultrastructural synaptic parameters

pharmacological behavioral designs, behavioral experiments for emotional behavior, learning and memory formation

in vivo microdialysis: quantitative measurements of amino acids, monoamines and their metabolites in freely behaving animals

cell cultures: organotypic cocultures of forebrain and thalamus, primary cultures of hippocampal and cortical neurons
